DESCRIPTION: Sildenafil is an inhibitor of phosphodiesterase 5 and is used as a treatment for erectile dysfunction in men and as a treatment for pulmonary hypertension.
PHARMACOLOGICAL ACTION: The physiological mechanism of penile erection involves the release of nitric oxide in the cavernous body during sexual stimulation, this then activates the enzyme guanylate cyclase, which increases the levels of cyclic guanosine monophosphate (cGMP), producing muscle relaxation smooth of the cavernous body and allowing the inflow of blood.
INDICATIONS AND USES: It is indicated as a coadjuvant in the management of erectile dysfunction.
DRUG INTERACTIONS: Sildenafil potentiates the hypotensive effects of nitrates and its concomitant use is contraindicated. The simultaneous use of α-blockers (terazocín, prazocín, doxazocín) can produce hypotension.
CONTRAINDICATIONS AND PRECAUTIONS: It is contraindicated in patients with known hypersensitivity to any of its components, patients with predisposition to pigmentosa retinopathy.
ADVERSE REACTIONS: In controlled clinical studies, the percentage of discontinuation due to secondary adverse events to sildenafil (2.5%) is not statistically significant with respect to the percentage of discontinuation found in the placebo group (2.3%). The incidence of some adverse effects increased in relation to the dose.
DOSAGE AND ROUTE OF ADMINISTRATION: It can be administered in a dose of 50 mg per day without exceeding 100 mg / day orally, one hour before sexual intercourse.

OVERDOSAGE: In studies with healthy volunteers and single doses of up to 800 mg, adverse events were similar to those observed with lower doses, but incidence rates and severities were increased.
